SPT6 Activators

SPT6 activators indirectly promote the functional activity of SPT6, a protein implicated in transcription elongation and chromatin remodeling. Forskolin, by boosting intracellular cAMP levels, and Rolipram, through inhibiting cAMP breakdown, both enhance PKA activity, which could phosphorylate partners involved in transcription, potentially aiding SPT6's role in elongation. Trichostatin A and BIX-01294, by inhibiting histone deacetylase and G9a histone methyltransferase respectively, lead to an open chromatin state, thereby facilitating SPT6's access to DNA and enhancing its elongation capability. Similarly, 5-Azacytidine decreases DNA methylation, which may complement the function of SPT6 in promoting transcriptional activity. Caffeine's antagonistic effect on adenosine receptors might culminate in a cellular state that favors transcriptional processes where SPT6 is active, due to elevated cAMP levels.

Further contributing to the activation of SPT6 are compounds that affect the stability and interaction of transcriptional machinery. Isoquercetin and (-)-Epigallocatechin gallate, by modulating topoisomerase IIactivity and kinase activity respectively, could alleviate tension during transcription elongation and shift kinase-mediated signaling in favor of SPT6's function. SRT1720, by activating SIRT1, may enhance SPT6's chromatin-related activities through deacetylation of transcription factors. Lastly, SR8278 antagonizes REV-ERB, which may lead to the enhancement of SPT6's role in elongation by modulating circadian rhythms of gene expression.
