In parallel, IBMX, by inhibiting phosphodiesterases, prevents the breakdown of cAMP, thus sustaining PKA activity and the subsequent phosphorylation of Smok2a. Epinephrine and Glucagon, both of which bind to their respective receptors, also cause an increase in cAMP concentration, further promoting the activation of PKA and subsequent phosphorylation of Smok2a. Dopamine, by interacting with dopaminergic receptors, can elevate cAMP in specific neuronal pathways, which activates PKA and leads to the phosphorylation of Smok2a. Anisomycin activates stress-activated protein kinases, which can phosphorylate Smok2a, while Phorbol 12-myristate 13-acetate (PMA) activates protein kinase C (PKC), which also has the capacity to phosphorylate and modulate Smok2a activity. Lastly, the introduction of a Nitric Oxide Donor like SNAP can lead to the activation of guanylate cyclase, increasing cGMP levels and potentially activating protein kinase G (PKG), which may then phosphorylate and activate Smok2a.
