RGS1 inhibitors, broadly speaking, include chemicals that interfere with the activation and inactivation cycle of G proteins. Pertussis Toxin and Cholera Toxin, for instance, ADP-ribosylate Gαi and Gαs subunits of G proteins, respectively. By modifying these G proteins, they prevent their interaction with G protein-coupled receptors (GPCRs), thereby indirectly affecting the function of RGS1, which acts as a GTPase-activating protein for these G proteins.
Similarly, YM-254890, NF023, UBO-QIC, and M119 specifically inhibit certain Gα proteins. YM-254890 is a specific inhibitor of Gαq/11 proteins, while NF023 is a non-competitive antagonist of Gαs and Gαi/o proteins. UBO-QIC and M119 selectively inhibit Gαi proteins. By preventing the activation of these G proteins, these compounds indirectly affect the function of RGS1. Gallein, on the other hand, inhibits Gβγ signaling. As Gβγ subunits are part of the heterotrimeric G proteins that RGS1 interacts with, inhibiting Gβγ signaling could indirectly affect RGS1. Lastly, MRS2179, Suramin, TNP-ATP, PPADS, and Reactive Blue 2 are all antagonists of P2 receptors, which are a class of GPCRs. By inhibiting the activation of G proteins via these receptors, these compounds could indirectly affect the function of RGS1.
