plexin-C1 Activators

Plexin-C1 Activators are a class of compounds that enhance the function of the plexin-C1 protein through diverse mechanisms, primarily via activation of specific signaling pathways that lead to increased ubiquitination processes. These compounds range from small organic molecules such as anisomycin, etoposide, and sorbitol, to biological molecules like epidermal growth factor (EGF), insulin, and tumor necrosis factor alpha (TNFα). Anisomycin, sodium arsenite, thapsigargin, and tunicamycin are potent activators of the JNK pathway, a key regulator of cell stress responses. Activating the JNK pathway can lead to phosphorylation of substrates involved in ubiquitination, thereby enhancing the function of plexin-C1 in signal transduction.

Meanwhile, Etoposide, a DNA damage response modulator, can activate the ATM/ATR pathway, leading to enhanced ubiquitination processes and increased plexin-C1 functionality. Paclitaxel, sorbitol, and hydrogen peroxide (H2O2) act by stabilizing microtubules and inducing osmotic and oxidative stress respectively, leading to activation of the MAPK pathway, a crucial route to ubiquitination. Compounds such as EGF, insulin, and TNFα trigger their respective receptor pathways – the EGFR, insulin receptor, and NF-κB pathways – which are interconnected with the MAPK pathway. This interconnection provides an alternative route to enhancing ubiquitination processes, and subsequently, plexin-C1 function. Lastly, U0126, a selective inhibitor of MEK1/2, indirectly activates upstream components of the MAPK pathway, leading to increased ubiquitination and enhanced plexin-C1 functionality.
