PGI2 synthase Activators are chemicals that can directly or indirectly stimulate the activity of PGI2 synthase, an enzyme that catalyzes the conversion of prostaglandin H2 (PGH2) to prostaglandin I2 (PGI2). These activators may act primarily on PGI2 synthase or they can target other elements of the prostaglandin synthesis pathway to increase PGI2 production. The chemicals listed above are primarily activators of the cyclooxygenase-2 (COX-2) enzyme, which is directly involved in the synthesis of PGI2. For instance, Aspirin acetylates COX-2, leading to a switch in enzyme activity to produce precursors for PGI2 synthesis. Similarly, Indomethacin and Resveratrol enhance COX-2 expression, and thereby, the initial steps of PGI2 synthesis.
Certain compounds, like Iloprost, Beraprost, Epoprostenol, and Treprostinil, are synthetic analogs of PGI2. These compounds do not directly activate PGI2 synthase but can stimulate endogenous PGI2 synthesis through feedback mechanisms. The presence of these PGI2 analogs in the cell can lead to increased production of endogenous PGI2. Cilostazol, on the other hand, increases intracellular cAMP levels, which can subsequently enhance COX-2 activity and PGI2 synthesis. Sulprostone, Misoprostol, and Enprostil increase endogenous PGI2 synthesis by binding to prostaglandin E2 receptors (EP3 receptors). This binding increases COX-2 activity, leading to enhanced synthesis of PGI2.
