PGI2 synthase Inhibitors are chemicals that can directly or indirectly inhibit the activity of PGI2 synthase, an enzyme that catalyzes the conversion of prostaglandin H2 (PGH2) to prostaglandin I2 (PGI2). These inhibitors may act primarily on PGI2 synthase or they can target other elements of the prostaglandin synthesis pathway to decrease PGI2 production.
The chemicals listed above are primarily inhibitors of the cyclooxygenase-2 (COX-2) enzyme, which is directly involved in the synthesis of PGI2. For instance, NS-398, Celecoxib, Etoricoxib, Parecoxib, Rofecoxib, Valdecoxib, Lumiracoxib, Deracoxib, Firocoxib, and Meloxicam are selective COX-2 inhibitors. These compounds reduce PGI2 synthesis by inhibiting the COX-2 enzyme and thereby reducing the production of prostaglandin precursors necessary for PGI2 synthesis. Aspirin and Indomethacin can also inhibit both COX-1 and COX-2 enzymes. At higher doses, these compounds can reduce PGI2 synthesis by inhibiting these enzymes and limiting the availability of precursor molecules.
