Chemical activators of Olr400 can engage various intracellular signaling pathways leading to its activation. Forskolin directly stimulates adenylyl cyclase, which catalyzes the conversion of ATP to cyclic AMP (cAMP). Elevated cAMP levels in turn activate protein kinase A (PKA), which is known to phosphorylate target proteins, thus Olr400 can be activated through this mechanism. Similar to forskolin, Isoproterenol functions as a beta-adrenergic agonist to elevate cAMP levels, again leading to PKA activation and potentially culminating in the phosphorylation and activation of Olr400. Histamine, via its action on H2 receptors, and Prostaglandin E2, through EP2/EP4 receptors, both increase cAMP in their target cells. This rise in cAMP can lead to the activation of PKA, which is a kinase that can phosphorylate Olr400, thereby activating it. Glucagon, a hormone, also binds to its specific receptor to cause an increase in cAMP and subsequent activation of PKA, which may then activate Olr400 by phosphorylation.
Inhibition of cAMP breakdown also leads to Olr400 activation. IBMX, Vardenafil, and Zaprinast all act as phosphodiesterase inhibitors, preventing the degradation of cAMP, thus leading to sustained PKA activity and subsequent phosphorylation and activation of Olr400. Adenosine operates through A2 adenosine receptors, again raising the cAMP levels, thus PKA can phosphorylate and activate Olr400. Cholera toxin persistently activates the Gs alpha subunit, which then continuously stimulates adenylyl cyclase, leading to an increase in cAMP, PKA activation, and the activation of Olr400. PACAP, a pituitary adenylate cyclase-activating peptide, binds to its receptors and increases cAMP levels, leading to PKA activation and, hence, Olr400 activation. Lastly, Anisomycin activates stress-activated protein kinases, which can also phosphorylate and activate Olr400, albeit through a different signaling mechanism than the cAMP-PKA axis. All these chemicals, through various pathways, ensure the activation of Olr400 by influencing the cellular environment and signaling cascades that lead to its phosphorylation and functional activation.
