Nuclear Factor of Activated T-cells (NFAT) represents a family of transcription factors involved in the regulation of gene expression in response to cellular stimuli. The NFAT family is comprised of multiple isoforms, one of which is NFATc3. This isoform, like other members of the NFAT family, plays a pivotal role in transducing intracellular calcium signals into changes in gene expression. Upon an increase in intracellular calcium concentration, the phosphatase calcineurin is activated, which subsequently dephosphorylates NFAT proteins. This dephosphorylation allows NFATc3 to translocate from the cytosol to the nucleus, where it can bind to specific DNA sequences and regulate the transcription of target genes. NFATc3 is known to participate in various cellular processes, including immune responses, muscle development, and cardiac functioning.
NFATc3 inhibitors are compounds specifically designed to modulate the activity of the NFATc3 transcription factor. These inhibitors operate by either preventing the activation and nuclear translocation of NFATc3 or by hindering its ability to bind to DNA and regulate gene expression. The mechanisms of action can be diverse, ranging from direct binding to the NFATc3 protein, inhibiting the upstream activator calcineurin, or altering post-translational modifications that affect NFATc3 activity.
