NDUFA7 Inhibitors

NDUFA7, is the largest and first enzyme of the mitochondrial respiratory chain. When the action of Complex I is impeded by these compounds, electron transfer from NADH to ubiquinone is obstructed, which results in an inability to create the electrochemical gradient essential for ATP synthesis. Rotenone and Piericidin A are examples of classic Complex I inhibitors. They act by binding to the complex and inhibiting the transfer of electrons, thus affecting the overall electron transport chain. On the other hand, chemicals like Caprylic acid can act by uncoupling oxidative phosphorylation, which also leads to a reduced activity of Complex I. Another interesting compound is MPP+, a neurotoxin, which has been studied for its effects on the mitochondrial respiratory chain, particularly on Complex I. Compounds like Fluvastatin may indirectly affect Complex I, given that statins can influence mitochondrial function. It's noteworthy that the exact mechanism of inhibition can vary, and not all these compounds target the same site or function within Complex I. However, by inhibiting Complex I, they can indirectly affect the function of its subunits, including NDUFA7.
