NDST Activators

N-Deacetylase/N-Sulfotransferase 1 (NDST1) is an integral enzyme in the biosynthesis of heparan sulfate (HS), a sulfated polysaccharide that is a major component of the extracellular matrix and cell surface. NDST1 is responsible for the initial modification steps of HS chains, which are essential for creating specific binding sites for a variety of proteins, including growth factors, morphogens, and adhesion molecules.

NDST1 enzymatically removes acetyl groups from N-acetylglucosamine residues in the HS precursor and adds sulfate groups to the resulting free amino groups. This dual activity is crucial for the subsequent pattern of sulfation that dictates the functional specificity of HS chains. The pattern of sulfation is a key determinant of the binding affinity of HS for various ligands, influencing a multitude of biological processes such as cell signaling, cell proliferation, angiogenesis, and wound healing.The expression of NDST1 is regulated by factors that control cellular differentiation and development. Aberrant expression or function of NDST1 is implicated in several pathological conditions, including cancer, where altered HS structure can affect tumor growth and metastasis.
