NDST Antibody (E-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects NDST protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). NDST (E-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NDST protein plays a crucial role in the modification of heparan sulfate and heparin, specifically through the N-deacetylation and N-sulfation of N-acetylglucosamine residues. This modification is essential for the synthesis of oligosaccharide sequences that possess specific ligand-binding properties, which are vital for various biological processes, including cell signaling, growth factor activity, and the regulation of cell adhesion. NDSTs′ enzymatic activity is mediated by monomeric enzymes that exhibit two catalytic functions, and multiple isozymes of NDST have been identified, each with distinct tissue distributions and enzymatic characteristics. Phylogenetic studies indicate that the NDST family, comprising NDST1-4, evolved from a common ancestral gene, leading to the emergence of two subtypes, NDST1/2 and NDST3/4. NDST1, located on human chromosome 5q32-q33.1, shares significant homology with NDST2, which is found on chromosome 10q22, while NDST3 and NDST4 are closely linked on chromosome 4, mapping to 4q25-26 and 4q26-27, respectively. Expression patterns of NDST3 and NDST4 mRNA in various mouse tissues are more restricted compared to the abundant and ubiquitous expression of NDST1 and NDST2, highlighting the functional diversity and regulatory mechanisms of these enzymes in different biological contexts.

Can NDST (E-9): sc-374529 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: TinTin
Thank you for your question. Yes, NDST (E-9): sc-374529 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
