NALP4B Activators

Chemical activators of NALP4B include a variety of compounds that initiate its activation through different mechanistic pathways. ATP, a ubiquitous energy molecule, directly engages with NALP4B to facilitate its oligomerization, a critical step for the protein's activation. This process is fundamental to the function of NALP4B, as the formation of the inflammasome complex triggers downstream immune responses. Similarly, the calcium ionophore A23187 raises intracellular calcium levels, which is a well-established signal for activating NALP4B. Elevated calcium levels can act as a second messenger within the cell, leading to a cascade of events that culminate in the activation of the inflammasome. On the other hand, nigericin disrupts potassium ion balance within the cell, and since intracellular potassium levels are tightly linked with inflammasome activation, its reduction directly leads to the activation of NALP4B.

Furthermore, aluminum potassium sulfate promotes conditions that favor the assembly of the NALP4B inflammasome, thereby triggering its activation. Monosodium urate (MSU) crystals are known to physically engage with components of the inflammasome, causing a structural rearrangement that is essential for NALP4B activation. Imiquimod, through its action on toll-like receptors, leads to the production of reactive oxygen species (ROS), another signaling molecule known to activate NALP4B. Muramyl dipeptide and lipopolysaccharide (LPS) both function through their interactions with NOD-like and toll-like receptors, respectively, which can then facilitate the activation of NALP4B by enhancing the signaling pathways that lead to inflammasome assembly.
