Similarly, CRID3 also inhibits NALP4B's ATPase activity, leading to a failure in the proper activation of this inflammasome complex. Another chemical, Parthenolide, interacts with NALP4B by modifying cysteine residues within its NACHT domain, which is essential for the oligomerization process that is a prerequisite for NALP4B's biological activity. Bay 11-7082 takes a different approach; it interferes with the ubiquitination that is pivotal for the activation and signaling of NALP4B, effectively preventing its normal inflammatory signaling cascade.
Further inhibitory actions include Glyburide and Belnacasan, which disrupt NALP4B function by targeting the NLRP inflammasome and caspase-1, respectively.
