MyD88 activators are a distinct class of chemicals that play a pivotal role in modulating immune response by indirectly influencing the activation of the MyD88 protein, coded by the MYD88 gene. These chemicals interact with various Toll-like receptors (TLRs) and effectively cause the recruitment of MyD88, facilitating downstream signaling cascades. The modus operandi of these activators spans diverse agonistic interactions with TLRs. For example, Poly(I:C) triggers TLR3, involving MyD88 in downstream activities. Imiquimod and Resiquimod target TLR7, with the latter also having activity on TLR8, and both result in MyD88-dependent signaling. On the other hand, Pam3CSK4 stimulates TLR1/2, which then induces the recruitment of MyD88 for subsequent signaling cascades.
The other half of this chemical cohort-Zymosan, R848, MALP-2, CL097, and Tri-DAP-also show similar versatility in receptor targeting and consequent MyD88 activation. Zymosan primarily influences TLR2 but also involves MyD88 in the downstream responses. Flagellin engages TLR5, leading to MyD88-dependent signaling. R848 acts on both TLR7 and TLR8 and initiates MyD88-dependent pathways. MALP-2 triggers TLR2/6 and recruits MyD88. CL097 targets TLR7/8 and results in the recruitment of MyD88 for downstream activities. Finally, Tri-DAP has a specialized role in engaging TLR2/1 heterodimers, initiating the recruitment of MyD88 for downstream immune responses.
