Mps1 (Monopolar spindle 1), also known as TTK, is a protein kinase crucial for ensuring accurate chromosome segregation during cell division. It is a core component of the spindle assembly checkpoint (SAC), a surveillance mechanism that blocks cells from dividing until all chromosomes are properly attached to the mitotic spindle.Mps1 functions primarily by phosphorylating various target proteins involved in the SAC, including kinetochore proteins, thereby controlling the attachment of the spindle microtubules to the kinetochores (structures on chromosomes where the spindle fibers attach during cell division). If chromosomes are not correctly attached, Mps1 inhibits the cell from progressing into anaphase, the stage of cell division where the chromosomes are separated.
Mps1 is also involved in the centrosome duplication cycle. It plays a role in initiating centrosome replication and inhibits overduplication, thus ensuring cells have the correct number of centrosomes.Aberrant Mps1 function can lead to chromosomal instability, a hallmark of many cancers. Overexpression of Mps1 has been observed in various types of cancer, including breast, lung, and colorectal cancers. Due to its role in cell division and its association with cancer, Mps1 has become a potential target for cancer.
