Dasatinib, for instance, targets the Src-family kinases which associate with immunoreceptor tyrosine-based activation motifs (ITAMs) that are key to MAIR-II signaling. By inhibiting these kinases, Dasatinib disrupts the activation signaling of MAIR-II. Similarly, PP2 and PD173955 function as Src-family tyrosine kinase inhibitors, obstructing the phosphorylation events that MAIR-II relies upon, thereby diminishing its signal transduction capabilities. Another inhibitor, SU6656, selectively inhibits Src family kinases, preventing the initiation of signaling cascades that facilitate MAIR-II function. AZD0530 and Bosutinib, both Src/Abl kinase inhibitors, work by blocking the phosphorylation of Src-family kinases, which is a prerequisite for downstream signaling involving MAIR-II.
Further inhibitory effects are observed with PP1, which selectively prevents Src family tyrosine kinases from phosphorylating key proteins in the MAIR-II signaling pathway. Saracatinib, another Src kinase inhibitor, impedes the activation of MAIR-II-associated pathways. SKI-606, which is also known as Bosutinib, serves a similar purpose by obstructing the kinase activity essential for MAIR-II's functional expression. KX2-391, by targeting the peptide substrate binding site of Src kinases, inhibits the critical kinases for MAIR-II signaling. Lastly, GNF-2, an allosteric Bcr-Abl inhibitor, affects Abl-related kinases and, in doing so, can disrupt the signaling pathways activated by MAIR-II, leading to its functional inhibition. Each of these chemicals operates by directly obstructing the specific kinases or phosphorylation processes that are fundamental for the activity of MAIR-II, resulting in its effective inhibition.
