Forskolin, a diterpene that exerts its effects by directly stimulating adenylyl cyclase, thereby ramping up the production of cAMP, a pivotal second messenger within the cell. This elevation of cAMP can lead to a series of phosphorylation events if the protein in question is regulated by cAMP-dependent pathways. IBMX, a methylxanthine derivative, which sustains elevated levels of cAMP by inhibiting phosphodiesterases, thus ensuring that any cAMP-driven processes are prolonged. This includes the activation of proteins regulated by phosphorylation under the governance of cAMP levels. Phorbol esters, typified by PMA, offer another route to protein activation through the stimulation of protein kinase C (PKC). PKC activation can directly influence proteins that are sensitive to phosphorylation by serine and threonine kinases. Ionomycin, on the other hand, bypasses surface receptors and directly increases intracellular calcium levels, thus activating calcium-dependent proteins.

Growth factors such as PDGF, NGF, and EGF engage their respective receptors and trigger kinase cascades, which may culminate in the activation of proteins positioned within these pathways. Compounds like dibutyryl-cAMP, a cell-permeable analog of cAMP, serve to mimic the second messenger, thus directly activating proteins regulated by cAMP-dependent pathways. Conversely, calyculin A and staurosporine, although typically recognized as inhibitors for protein phosphatases and kinases respectively, can paradoxically lead to the activation of certain proteins by modulating the balance of phosphorylation within the cell.
