LCE1 Activators

Chemical activators of LCE1 operate through diverse mechanisms to enhance its function within cellular pathways. Calcium Chloride, for example, raises intracellular calcium levels, which then activate calcium-dependent protein kinases. These kinases can phosphorylate LCE1, an action that is essential for the protein's activation. Similarly, Ionomycin functions as a calcium ionophore, also raising intracellular calcium and activating kinases that phosphorylate and activate LCE1. Phorbol 12-myristate 13-acetate (PMA) and 12-O-Tetradecanoylphorbol-13-acetate (TPA) are both activators of protein kinase C (PKC). Once PKC is activated, it can directly phosphorylate LCE1, leading to its activation.

In the realm of phosphatase inhibition, Calyculin A and Okadaic Acid play significant roles. By inhibiting protein phosphatases, these chemicals maintain LCE1 in a phosphorylated state, which is indicative of its active form. Forskolin and Dibutyryl-cyclic AMP elevate cAMP levels, which in turn activate protein kinase A (PKA). PKA then phosphorylates LCE1, leading to its activation. Thapsigargin exerts its effect by inhibiting the SERCA pump, which causes an increase in intracellular calcium that activates kinases capable of phosphorylating LCE1. Phosphatidic Acid, through the activation of the mTOR signaling pathway, and Epidermal Growth Factor (EGF), through its receptor's downstream signaling pathways, both result in kinase activation, which can then phosphorylate and activate LCE1. Lastly, Anisomycin activates stress-activated protein kinases, such as JNK, which in turn may lead to the phosphorylation and subsequent activation of LCE1.
