IER3IP1, or Immediate Early Response 3 Interacting Protein 1, is a protein that in humans is encoded by the IER3IP1 gene. It is a transmembrane protein localized in the endoplasmic reticulum (ER) and is involved in the regulation of cellular stress responses, particularly in the context of the unfolded protein response (UPR).The UPR is a cellular stress response related to the ER. It is activated in response to an accumulation of unfolded or misfolded proteins in the ER lumen, which is a condition known as ER stress. The UPR aims to restore normal function by halting protein translation, degrading misfolded proteins, and activating the signaling pathways that lead to increased production of molecular chaperones involved in protein folding.
IER3IP1 has been shown to interact with components of the UPR signaling pathway, and its expression is induced by various forms of ER stress. It is believed to modulate the UPR and may act as a regulator of apoptosis, as dysregulation of ER homeostasis can lead to cell death. This is particularly important in maintaining the health of secretory cells that are involved in intensive protein synthesis and folding, such as pancreatic beta cells and neurons.Mutations in the IER3IP1 gene have been associated with a rare form of autosomal recessive microcephaly with epileptic encephalopathy, suggesting its crucial role in brain development and function. This phenotype indicates that IER3IP1 may be necessary for the proper development and survival of neural cells, which are particularly sensitive to disturbances in protein folding and ER stress.
