Heparan-alpha-glucosaminide N-acetyltransferase (HGSNAT) is a protein that in humans is encoded by the HGSNAT gene. This protein plays a critical role in the degradation of heparan sulfate, a type of complex sugar molecule, within the lysosome. The lysosome is a cellular organelle involved in the breakdown and recycling of various biomolecules.HGSNAT catalyzes the acetylation step in heparan sulfate degradation, which involves the transfer of an acetyl group to the sugar molecule. This modification is essential for the subsequent degradation of heparan sulfate.
Mutations in the HGSNAT gene can disrupt the normal function of the HGSNAT protein, leading to the accumulation of undegraded heparan sulfate in the lysosomes. This leads to a group of disorders known as mucopolysaccharidoses, specifically Mucopolysaccharidosis type IIIC (also known as Sanfilippo Syndrome type C).Sanfilippo Syndrome is a severe neurodegenerative disorder that typically presents in childhood.indicators include developmental delay, behavioral problems, and progressive intellectual decline. The disease is currently incurable, often do not survive beyond adolescence or early adulthood. The HGSNAT gene is widely expressed in various tissues, reflecting the importance of heparan sulfate degradation in normal physiological function. However, the impact of HGSNAT dysfunction seems to be most severe in the nervous system.
