GPR175 Activators

GPR175 Activators can be divided into several categories. The first group includes Anandamide and Cannabidiol. These cannabinoids can enhance GPR175 activation via their interaction with G-protein coupled receptors (GPCRs), thereby amplifying GPCR signaling events. Anandamide and Cannabidiol stimulate cannabinoid receptors, which share signaling pathways with GPR175, leading to enhanced GPR175 activation via G-protein mediated signaling. Similarly, Capsaicin, a vanilloid compound that acts as a ligand for transient receptor potential vanilloid 1 (TRPV1), can trigger GPCR activation and potentially enhance GPR175 activity through G-protein mediated signaling events.

Another group of GPR175 activators includes Resveratrol and Curcumin. Resveratrol, a polyphenolic compound, activates sirtuins, which can affect the activity of GPCRs, thereby enhancing the functional activity of GPR175. Likewise, Curcumin, another polyphenolic compound, can enhance GPR175 activation by affecting the G-protein mediated signaling pathways that GPR175 is directly involved in. Forskolin, Quinidine, Epinephrine, Dopamine, Acetylcholine, L-Arginine, and Sildenafil form another category of GPR175 activators. These compounds act on different targets, but ultimately converge on GPCR signaling pathways, enhancing GPR175 function. For example, Forskolin directly activates adenylyl cyclase, leading to an increase in intracellular cAMP levels and subsequent enhancement of GPR175 activity. Similarly, L-Arginine and Sildenafil enhance GPR175 function by manipulating cGMP levels, a pathway in which GPR175 is known to be involved.

L-Arginine is a substrate for nitric oxide synthase (NOS), which produces nitric oxide (NO). NO can activate guanylate cyclase, leading to increased cGMP levels. The elevated cGMP levels can enhance GPR175 function because GPR175 is involved in cGMP-dependent signaling pathways.
