GPR148 Inhibitors

Pertussis Toxin and Cholera Toxin have opposing effects on G protein signaling; the former inhibits the Gi/o type G proteins, potentially reducing GPR148 mediated inhibitory effects, while the latter permanently activates Gs proteins, which could override GPR148's signaling. Forskolin, by directly activating adenylyl cyclase, can increase intracellular cAMP levels, which may modulate cellular responses regulated by GPR148 or bypass its regulatory effects entirely. Propranolol and Losartan interfere with GPCR signaling by blocking beta-adrenergic and angiotensin II receptors, respectively. These antagonists can prevent receptor-mediated responses within signaling networks that GPR148 may participate in. Y-27632 and ML7 target the cytoskeletal regulatory proteins ROCK and myosin light chain kinase, respectively, affecting cellular processes that could be regulated by GPR148. PD98059 and GW5074 interfere with the MAPK/ERK pathway at different points, potentially influencing downstream signaling of GPR148, which may be implicated in cell proliferation and differentiation. LY294002 targets the PI3K/Akt pathway, a critical signaling cascade involved in a variety of cellular functions, which GPR148 may influence. Go 6983 and U73122 disrupt protein kinase C and phospholipase C activities, respectively, which are important components of GPCR signaling.
