fringe Activators

Fringe activators are a class of compounds that enhance the expression or activity of the Fringe protein. Fringe is a glycosyltransferase enzyme that plays a crucial role in the Notch signaling pathway, which is involved in various cellular processes, including cell fate determination, differentiation, proliferation, and apoptosis.

Fringe activators exert their effect by modulating different aspects of Fringe biology. Some Fringe activators, such as lithium chloride and Wnt3a, act upstream of Fringe by activating the canonical Wnt signaling pathway, which leads to the stabilization of β-catenin and increased Fringe expression. Other Fringe activators, such as CHIR99021 and IWR-1, directly target and inhibit GSK-3, a kinase that phosphorylates and targets β-catenin for degradation. By inhibiting GSK-3, these compounds stabilize β-catenin and enhance Fringe expression.Other Fringe activators, such as BIO, LGK974, XAV939, C59, ICG-001, WNT974, PRI-724, and NVP-TNKS656, target the Tankyrase enzyme, which is involved in the degradation of β-catenin. By inhibiting Tankyrase, these compounds stabilize β-catenin and enhance Fringe expression.

By modulating Fringe expression or activity, Fringe activators can influence Notch signaling and impact various cellular processes. These compounds have been instrumental in dissecting the molecular mechanisms underlying Notch signaling and its involvement in various biological processes and disease pathogenesis.
