FBXO8 Inhibitors

FBXO8 inhibitors, as a chemical class, are not defined by a singular molecular structure or a shared pharmacophore. Instead, their unifying characteristic is the ability to modulate the activity of the FBXO8 protein indirectly, primarily through interference with the ubiquitin-proteasome system (UPS) and related cellular processes. These inhibitors are usually broader-acting proteasome inhibitors or molecules that affect upstream processes that dictate substrate ubiquitination and subsequent proteasomal degradation. FBXO8 inhibitors can stabilize proteins targeted for degradation. By doing so, they can inadvertently affect the turnover of substrates that FBXO8 might tag for proteolysis. Additionaly, FBXO8 inhibtors act on NEDD8-activating enzyme, which is crucial for the neddylation process-a post-translational modification that can regulate the activity of the SCF complex, of which FBXO8 is a component. These inhibitors represent compounds that act on stress response pathways and autophagy, respectively, both of which can have downstream effects on protein degradation and turnover. This illustrates the indirect approach necessary to target FBXO8, involving a wide array of biochemical pathways and cellular processes. The diversity of these compounds underscores the multifaceted nature of protein regulation in the cell and the challenges in specifically modulating one component of this complex system.
