CST Inhibitors

CST inhibitors are chemical compounds that specifically target and inhibit the function of CST (CST complex), a protein complex that plays a critical role in telomere maintenance and DNA replication. The CST complex, composed of CTC1, STN1, and TEN1, is essential for protecting telomeres and facilitating the proper replication of chromosome ends. Telomeres, which are repetitive nucleotide sequences at the ends of chromosomes, prevent the loss of essential genetic information during cell division. The CST complex ensures that telomeres are adequately replicated, particularly by coordinating the final steps of telomere replication, where traditional replication machinery struggles to complete the process. CST acts as a single-strand DNA-binding protein, stabilizing the telomere ends and preventing them from being recognized as damaged DNA.

Inhibitors of CST disrupt the function of the complex by blocking its ability to bind to telomeric DNA or interfering with its interactions within the complex itself. These inhibitors may target specific binding domains within the CTC1, STN1, or TEN1 components, preventing the CST complex from performing its protective role at telomeres. By inhibiting CST, researchers can study its direct role in telomere length regulation and the broader implications of telomere stability on genome integrity. Disrupting CST function can lead to improper replication of telomeres, exposing the effects of telomere shortening and genomic instability, which are critical factors in cellular aging and DNA damage responses. CST inhibitors thus serve as important tools for investigating the cellular mechanisms that safeguard chromosome ends and maintain genetic stability during cell division.
