Ciprofloxacin Activators

Ciprofloxacin is a broad-spectrum fluoroquinolone antibiotic that exerts its bactericidal effects by inhibiting the activity of bacterial DNA gyrase and topoisomerase IV enzymes, both of which are essential for bacterial DNA replication, transcription, and repair. By targeting these enzymes, ciprofloxacin interferes with bacterial DNA synthesis and ultimately leads to bacterial cell death. Specifically, ciprofloxacin binds to the A subunit of DNA gyrase and the subunit B of topoisomerase IV, forming stable drug-enzyme-DNA complexes that prevent the resealing of DNA breaks generated during DNA replication. This results in the accumulation of double-strand breaks in bacterial DNA, leading to the activation of bacterial SOS response and subsequent cell death.

Upon administration, ciprofloxacin undergoes rapid absorption from the gastrointestinal tract and achieves concentrations in various tissues and body fluids. Once absorbed, ciprofloxacin penetrates bacterial cells through passive diffusion and active transport mechanisms. Within the bacterial cell, ciprofloxacin binds to its target enzymes, DNA gyrase, and topoisomerase IV, with high affinity, forming stable drug-enzyme complexes that prevent the proper functioning of these enzymes. The inhibition of DNA gyrase and topoisomerase IV disrupts the normal process of DNA replication and transcription, leading to the accumulation of DNA strand breaks and the activation of bacterial SOS response. Ultimately, the bactericidal effects of ciprofloxacin result from the interference with bacterial DNA synthesis and repair, leading to cell death.
