Chemical activators of ADP-ribosyltransferase 2b play a pivotal role in its enzymatic function. NAD+, as a primary substrate, directly participates in the ADP-ribosylation process that ADP-ribosyltransferase 2b catalyzes. The availability of NAD+ within the cell is crucial for the activation of ADP-ribosyltransferase 2b, as it provides the necessary ADP-ribose units that the enzyme transfers onto target proteins. Nicotinamide serves a dual purpose; while being a component of the NAD+ molecule, it also regenerates NAD+ when it is converted back into the active form, thereby maintaining a supply that ADP-ribosyltransferase 2b requires for its activity. Thymidine supports the cellular pool of NAD+ by facilitating DNA synthesis pathways, which indirectly ensures a constant supply of NAD+ for ADP-ribosyltransferase 2b's enzymatic actions. NGD+ acts as an alternate substrate to NAD+ for ADP-ribosyltransferase 2b, enabling the enzyme to carry out ADP-ribosylation even when NAD+ levels are not optimal.

Further supporting the activity of ADP-ribosyltransferase 2b, Benzamide, by inhibiting other NAD+ consuming enzymes, effectively increases the NAD+ availability for ADP-ribosyltransferase 2b. Meta-iodobenzylguanidine, though not a direct substrate, influences cellular NAD+ levels, thereby indirectly supporting the enzymatic action of ADP-ribosyltransferase 2b. Compounds like 3-Aminobenzamide inhibit poly(ADP-ribose) polymerases, which compete for NAD+ within the cell, thus raising the NAD+ levels available for ADP-ribosyltransferase 2b. Nicotinamide analogues, such as 6-Aminonicotinamide, increase cellular NAD+ pools, indirectly augmenting the activation of ADP-ribosyltransferase 2b. Nicotinic acid, another precursor to NAD+, contributes to the biosynthesis of NAD+ and consequently, to the activation of ADP-ribosyltransferase 2b. Similarly, Nicotinamide riboside and NMN (nicotinamide mononucleotide) are precursors that are converted into NAD+ within the cell, thereby facilitating the activation of ADP-ribosyltransferase 2b. Lastly, the inhibition of NRK1 can lead to increased levels of nicotinamide riboside, which, after conversion to NAD+, activates ADP-ribosyltransferase 2b by ensuring an adequate substrate supply for its ADP-ribosylation activity.
