APP Inhibitors

APP inhibitors belong to a chemical class of compounds designed to target amyloid precursor protein (APP), a transmembrane protein that plays a critical role in the pathogenesis of Alzheimer's disease. APP is a large, single-pass transmembrane protein found in various cell types, and it is proteolytically cleaved to produce amyloid-beta (Aβ) peptides. The abnormal accumulation and aggregation of these Aβ peptides are associated with the formation of amyloid plaques, one of the hallmarks of Alzheimer's disease. APP inhibitors act by interfering with the processing of APP, particularly by modulating the enzymes involved in its cleavage. The primary enzymes targeted by APP inhibitors are beta-secretase (BACE1) and gamma-secretase. BACE1 initiates the cleavage of APP, leading to the formation of a soluble extracellular fragment and a C-terminal fragment. Subsequently, gamma-secretase mediates the final cleavage step, releasing various Aβ isoforms, including Aβ40 and the more neurotoxic Aβ42.

The goal of APP inhibitors is to reduce the production of toxic Aβ aggregates by inhibiting the activity of BACE1 and/or gamma-secretase, thus attenuating the formation of amyloid plaques. Researchers are actively exploring various small molecules and biologics as APP inhibitors. However, the development of APP inhibitors is complex, as APP has other physiological functions, and interfering with its processing may have unintended consequences. The study of APP inhibitors is essential for advancing our understanding of the molecular mechanisms underlying Alzheimer's disease. By investigating how these inhibitors modulate APP processing, researchers hope to gain insights into the regulation of Aβ production and aggregation. This knowledge may pave the way for the development of novel strategies to combat Alzheimer's disease by targeting the early steps in Aβ generation and deposition.
