AFAP-1L1 Inhibitors

Chemical inhibitors of AFAP-1L1 function by targeting the cellular mechanisms that regulate the protein's activity, particularly its interaction with actin cytoskeleton dynamics. PP2, Dasatinib, SU6656, Bosutinib, Saracatinib, KX2-391, and Ponatinib are all Src family kinase inhibitors that inhibit the phosphorylation of AFAP-1L1. Phosphorylation by Src family kinases is a key regulatory step for AFAP-1L1, which, when phosphorylated, can associate with actin filaments and influence their organization. By preventing phosphorylation, these inhibitors reduce AFAP-1L1's ability to cross-link actin filaments, effectively inhibiting its function in the modulation of the actin cytoskeleton. SKI-606 (Bosutinib) operates similarly, with its dual inhibition of Src and Abl kinases, leading to a decrease in the phosphorylation of AFAP-1L1, which is necessary for its actin-binding activity.

The functional inhibition of AFAP-1L1 is further achieved through the action of PF-562271, a focal adhesion kinase (FAK) inhibitor. FAK is known to interact with AFAP-1L1, and its inhibition can disrupt this interaction, which is crucial for AFAP-1L1's role in actin filament organization. GNF-2, an allosteric inhibitor of Bcr-Abl that also targets Src kinases, inhibits the kinase activity required for AFAP-1L1's phosphorylation, thus diminishing AFAP-1L1's ability to affect actin dynamics. AZD0530 and PP1, both Src/Abl kinase inhibitors, similarly inhibit the phosphorylation of AFAP-1L1, preventing its role in organizing and stabilizing the actin cytoskeleton. The inhibition of these kinases results in a direct inhibition of AFAP-1L1's actin regulatory functions, thereby impeding its normal role in cytoskeletal arrangements.
