MAN1C1 Inhibitors

Chemical inhibitors of Alpha-mannosidase 2C1 (MAN1C1) primarily focus on disrupting the glycoprotein processing pathways in which this enzyme is involved. Swainsonine, an inhibitor of Golgi alpha-mannosidase II, impedes a similar process as MAN1C1, thereby potentially inhibiting its function in glycosylation. Kifunensine, targeting mannosidase I, can indirectly inhibit MAN1C1 by affecting the early stages of N-linked glycan processing. This impact on early glycosylation stages is also seen with Deoxymannojirimycin and Deoxynojirimycin, both of which are mannosidase inhibitors that can interfere with MAN1C1's role in glycoprotein processing. Similarly, Castanospermine and 1-Deoxynojirimycin, by inhibiting related enzymes in glycoprotein processing, could hinder the functional activity of MAN1C1, especially in N-linked glycan maturation.Further, Miglustat, by inhibiting glucosylceramide synthase, can indirectly affect MAN1C1's activity in glycolipid metabolism, thus impacting its role in glycoprotein processing. Alpha-glucosidase inhibitors like Voglibose and Acarbose can disrupt glycoprotein processing pathways, affecting N-linked glycan modification where MAN1C1 is active. Salbostatin, as a trehalase inhibitor, potentially influences similar glycoprotein processing pathways. Pyrrolidine dithiocarbamate, an NF-kB inhibitor, may also indirectly inhibit MAN1C1 by affecting signaling pathways that regulate glycoprotein processing. Lastly, Brefeldin A, which inhibits protein transport from the endoplasmic reticulum to the Golgi apparatus, could disrupt the pathway integral to MAN1C1's function. Each of these chemicals, through their respective targets, converges on the glycoprotein processing pathway, indirectly leading to the inhibition of MAN1C1's role in this essential cellular process.

Kifunensine acts as a selective inhibitor of the MAN1C1 enzyme, exhibiting distinctive molecular interactions that disrupt the hydrolysis of mannose-rich oligosaccharides. Its unique stereochemistry facilitates strong binding to the enzyme's active site, leading to altered reaction kinetics. This compound's interference with glycan remodeling processes highlights its role in modulating glycoprotein maturation and influencing cellular glycan profiles, thereby impacting various biological pathways.
