Chemical activators of Inka box actin regulator 1 can influence its activity through various biochemical pathways primarily by modulating the phosphorylation state of the protein. Phorbol 12-myristate 13-acetate (PMA), a potent activator of Protein Kinase C (PKC), plays a pivotal role by directly phosphorylating Inka box actin regulator 1, thereby enhancing its activity. Similarly, Forskolin, by elevating cAMP levels, activates Protein Kinase A (PKA), which can also target Inka box actin regulator 1 for phosphorylation, leading to its activation. Additionally, ionomycin, by increasing intracellular calcium levels, activates calmodulin-dependent kinases capable of phosphorylating Inka box actin regulator 1, which results in its activation.

Other chemicals such as Okadaic Acid and Calyculin A function by inhibiting protein phosphatases PP1 and PP2A, which prevents the dephosphorylation of proteins, including Inka box actin regulator 1, thus maintaining it in an active state. Epidermal Growth Factor (EGF) triggers its receptor, leading to the activation of the MAPK/ERK pathway, which can then activate Inka box actin regulator 1 through phosphorylation. Zinc Chloride can act as an allosteric modulator of kinase activity, potentially resulting in the phosphorylation and subsequent activation of Inka box actin regulator 1. Dibutyryl-cAMP, a cAMP analog, activates PKA, which is known to phosphorylate and activate Inka box actin regulator 1. Bisindolylmaleimide I and H-89 both inhibit PKC and PKA, respectively, but through feedback mechanisms can lead to the activation of alternative pathways that may phosphorylate and activate Inka box actin regulator 1. Anisomycin, by activating stress-activated protein kinases, contributes to the phosphorylation and activation of Inka box actin regulator 1. Lastly, Thapsigargin, through the elevation of intracellular calcium levels, can activate calcium-dependent kinases, which may in turn phosphorylate and lead to the activation of Inka box actin regulator 1, thereby modulating actin dynamics within the cell.
