GTBP Antibody (E-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects GTBP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). GTBP (E-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. GTBP, also known as MSH6, plays a crucial role in DNA mismatch repair pathway, which maintains genomic stability by correcting DNA replication errors. GTBP is primarily located in the nucleus, where GTBP forms a heterodimer with MSH2 to recognize and bind to G/T mismatches in DNA. This interaction initiates a cascade of events leading to excision and resynthesis of erroneous DNA segments. GTBP′s importance in this pathway is underscored by GTBP′s association with hereditary nonpolyposis colorectal cancer (HNPCC), where mutations in mismatch repair genes lead to microsatellite instability and increased cancer risk. Using anti-GTBP antibody (E-8) in research can provide valuable insights into DNA repair mechanisms and their dysfunction in cancer biology.

Can GTBP (E-8): sc-137015 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, GTBP (E-8): sc-137015 has been validated for use in IHC with paraffin-embedded sections. Rated 5 out of 5 by from Great featureI requested this antibody for free a couple weeks ago, the first time I run WB as 1:100, it works well, but has some non-specific bands. After that, the antibody works fine, one specific band appears around 180kD in my Hela cell samples in shorter exposure time.

Rated 3 out of 5 by from GTBP/Msh6 antibody (E-8) in Western blottingI received a sample antibody and tried it at a 1:100 dilution to probe whole cell extracts from various cell lines as labelled in my picture (note that GTBP runs at ~160 kDa). While it did not work in Mouse Embryonic Fibroblast (MEF) cells, it did work in human cell lines like HeLa, HEK293, and HEK293T (also known as Phoenix cells). There were some non-specific bands from this antibody, as seen in my western blot. If I were to buy this antibody or receive another sample, I might try a lower concentration next time. Overall, I'd say it's not bad.
