Fatty Acid 抗体

脂肪酸合成酶抗体（G-11）是一种小鼠单克隆IgG1 kappa轻链抗体，可通过蛋白质印迹（WB）、免疫沉淀（IP）、免疫荧光（IF）、免疫组织化学和酶联免疫吸附测定（ELISA）检测小鼠、大鼠和人类来源的脂肪酸合成酶（FASN）。FASN抗体（G-11）有非结合型和多种结合型，包括琼脂糖、辣根过氧化物酶（HRP）、藻红蛋白（PE）、异硫氰酸荧光素（FITC）和多种Alexa Fluor®结合物。FASN在脂质代谢中起着至关重要的作用，特别是在长链饱和脂肪酸的合成中，这些脂肪酸对于细胞膜的完整性和能量储存至关重要。FASN以同二聚体的形式存在，含有七个独特的催化位点，利用NADPH作为辅助因子，促进乙酰辅酶A和丙二酰辅酶A转化为脂肪酸。人类FASN基因编码的蛋白质由2504个氨基酸组成，在不同组织中的表达水平各不相同，在脑、肺和肝中的表达水平尤其高。值得注意的是，肿瘤细胞中经常出现脂肪酸合成失调的现象，其中FASN可能有助于恶性癌症中细胞的生长和存活，因此FASN成为治疗干预的潜在靶点。

Can Fatty Acid Synthase (G-11): sc-48357 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Fatty Acid Synthase (G-11): sc-48357 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Application: Western blot Sample: whole lysate of human hepatocytes from Huh7 cell line Gel running conditions: reduced, denaturing, 4-12% SDS-PAGE Loading amount: 30 µg protein/lane Transfer: semi-dry, on nitrocellulose membrane (0.45 µm) Blocking agent: 5% milk in TBST buffer, for 1h, RT Primary antibody dilution- 1: 400 in 5% milk, for 16 h at 4°C Secondary antibody: non- Santa Cruz antibody anti-mouse-HRP, 1:10000 dilution in 5% milk, for 1h, RT Detection: ECL substrate (femto sensitivity) Exposure time: 4 sec Bands: a strong, specific band at 260-270 kDa; non-specific bands between 130-180 kDa.
