ZO-1 Antibody (R40.76) is a rat monoclonal IgG2a antibody designed to detect the Zonula Occludens-1 protein, also known as Tight Junction Protein 1 (TJP1), in mouse, rat, and human samples using western blotting (WB), immunoprecipitation (IP), and PAR assays. Anti-ZO-1 antibody (R40.76) is raised against DOC-insoluble junctional ribbons isolated from rat liver, ensuring reliable recognition of the ZO-1 epitope. ZO-1 is a critical component of tight junctions, which are essential for maintaining the integrity and selective permeability of epithelial and endothelial cell layers. As a member of the membrane-associated guanylate kinase (MAGUK) family, ZO-1 interacts with other tight junction proteins such as occludin, claudins, ZO-2, and ZO-3 through its PDZ, SH3, and guanylate kinase domains, facilitating both structural stability and signal transduction. These interactions link the tight junction complexes to the actin cytoskeleton, playing a vital role in regulating cell polarity, barrier function, and cellular communication. Dysregulation of ZO-1 has been associated with various pathological conditions, including cancer metastasis, where ZO-1 expression is notably reduced in many breast cancer cell lines, as well as in cardiovascular and neurological disorders. Anti-ZO-1 antibody (R40.76) is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and Alexa Fluor® conjugates, providing versatile options for different experimental applications. ZO-1 monoclonal antibody (R40.76) enables researchers to investigate the expression, localization, and functional dynamics of ZO-1, thereby advancing our understanding of cellular junctions and their role in health and disease.

请问sc-33725(ZO-1)被不小心放进负二十度的冰箱里冻了一年,还可以再用吗?

感谢您的提问。sc-33725应在4°C保存,不可以冷冻。冷冻后的抗体实验效果无法得到保证。 Thank you for your question. sc-33725 should be stored at 4°C, don't freeze. There is no warranty after the antibody is forzen.

Não conseguimos detectar a banda refente ao ZO-1 (225kD). Estou enviando as imagens do "melhor" experimento que conseguimos até agora. Note que as bandas de beta-actina (42kD) estão ok, então acreditamos que não seja problema das nossas amostras uma vez que outros anticorpos,como E-caderina, estão funcionando normalmente. Gostaria de saber como podemos melhorar nosso protocolo de western blot. Obrigada

Rated 3 out of 5 by from Not great for IF as a plasma membrane markerThe antibody was used for immunofluorescence with secondary anti-rat antibody fused with Alexa647, in osteosarcoma cell lines U2OS and Saos2 and in WI-38 VA13 immortalized cell line. In U2OS and WI-38 VA13, the observed signal was not corresponding to the expected pattern, appearing as strong background with many faint foci, with most of the signal in the nucleus. In Saos2 cells, however, the signal was predominantly localized to intercellular boundaries, as expected, and in these cells, in our hands, this antibody could be used as a reliable marker of plasma membrane.
