XPA Antibody (2A4) is a mouse monoclonal IgG1 antibody that detects Xeroderma pigmentosum group A (XPA) in human samples through western blotting (WB). XPA plays a crucial role in the nucleotide excision repair (NER) pathway, which is essential for the cellular response to DNA damage caused by ultraviolet (UV) radiation. XPA protein is vital for maintaining genomic stability, as XPA coordinates the assembly of a preincision complex necessary for the repair of damaged DNA. XPA′s ability to recognize and bind to DNA is influenced by the structural integrity of the DNA double helix and the presence of bulky adducts, making XPA a key player in the repair process. Notably, XPA exists as a homodimer in the absence of DNA, but XPA′s binding efficiency increases significantly when interacting with DNA, highlighting the importance of its dimeric form in effective DNA repair. Deficiencies in XPA lead to an inability to repair UV-induced DNA damage, resulting in a heightened risk of skin cancers in affected individuals.
