
VPS28 Antibody (E-10) is a mouse monoclonal IgG2a antibody that detects vacuolar protein sorting protein 28 (VPS28) in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). VPS28 plays a crucial role in endocytic and biosynthetic trafficking processes essential for delivering proteins to the vacuole, a vital organelle involved in cellular waste processing and nutrient storage. Mutations in VPS28 can lead to accumulation of vacuolar endocytic and late Golgi markers within aberrant endosome-like class E compartments, disrupting normal cellular trafficking. These class E compartments contain endocytosed markers, precursors of vacuolar hydrolases, and markers typically associated with the trans-Golgi network, highlighting VPS28′s role in sorting materials destined for the vacuole away from those that cycle through the endocytic system. VPS28, along with other class E VPS proteins, facilitates formation of transport intermediates necessary for efficient transport out of the prevacuolar endosome, underscoring its importance in maintaining cellular homeostasis. VPS28 is localized in the cytoplasm and functions as a subunit of the endosomal sorting complex required for transport 1 (ESCRT-1), which is critical for protein sorting and transport within the cell.
