VMAT 1 Antibody (G-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects VMAT 1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). VMAT 1 (G-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. VMAT 1 protein, also known as SLC18A1, plays a crucial role in neurotransmission by facilitating the packaging of monoamines into synaptic vesicles, which is essential for the regulated release of neurotransmitters in neurons and neuroendocrine cells. This process is vital for proper neuronal communication and overall brain function, as it ensures that neurotransmitters are stored and released in a controlled manner, preventing excessive signaling that could lead to neurotoxicity. VMAT 1 structure is characterized by twelve transmembrane domains, which are integral to its function as a vesicular transporter, and shares weak homology with a class of bacterial antibiotic resistance proteins, highlighting its evolutionary significance. The vesicular transport mechanism employed by VMAT 1 is both ATP- and proton-dependent, underscoring the energy requirements for effective neurotransmitter packaging and release.

Is this antibody suitable for cell surface staining using flow cytometry? Was this tested?

Thank you for your question. Anti-VMAT 1 Antibody (G-12) is recommended for detection of VMAT 1 of mouse, rat and human origin by WB, IP, IF and ELISA. This antibody was raised against amino acids 44-143 of human protein, which corresponds to a Lumenal/vesicle topological domain. Fluorescent conjugates can be used for flow cytometry use, but permeabilization of the cells would be required.

What is the recommended fixation method for immunofluorescence staining with VMAT 1 (G-12): sc-166391 monoclonal antibody?
