Vimentin Antibody (3CB2) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 419-438 of human vimentin, a 57 kDa intermediate filament protein crucial for cellular architecture and function in mesenchymal cells. Vimentin mouse monoclonal antibody (3CB2) recognizes an epitope within the vimentin immunogen sequence. The molecular architecture of vimentin reveals sophisticated organization: vimentin molecules form α-helical coiled-coil dimers that assemble into higher-order structures, creating a stable yet dynamic filament system. This complex network serves multiple essential functions within cells - vimentin filaments maintain cell shape, position organelles, and enable cells to withstand mechanical stress. While vimentin predominantly resides in the cytoplasm, vimentin redistributes to the perinuclear region during cell division and stress conditions, facilitating processes such as cell migration, division, and adhesion. Vimentin participates in several crucial protein partnerships that extend beyond structural support. One significant interaction occurs with integrin-linked kinase (ILK), a key mediator of integrin signaling that controls cell adhesion, migration, and survival. Through vimentin′s association with ILK, cells can effectively transmit extracellular signals to the cytoskeleton - a process particularly important during epithelial-mesenchymal transition (EMT), when cells must extensively remodel their architecture for cancer progression and metastasis. Another critical partnership exists between vimentin and plectin, a large cytolinker protein that connects vimentin filaments to other cytoskeletal components such as actin filaments and microtubules. This interconnected network provides enhanced structural integrity, allowing cells to better resist mechanical forces, especially in tissues subject to significant physical stress like muscle and connective tissues. These sophisticated molecular interactions highlight vimentin′s role not only as a structural protein but as a central coordinator of cellular mechanics and signaling.

Is there any reactivity with lower vertebrates, such as frog or salamanders?

Thank you for your question! Anti-Vimentin Antibody (E-5) is recommended for detection of Vimentin of mouse, rat, human, canine and bovine origin by WB, IP, IF, IHC(P) and ELISA. There is no data supporting reactivity with samples from non-mammalian or amphibian species.

Is there data showing reactivity with canine samples?

Thank you for your inquiry. We do not have data showing reactivity of this antibody with canine samples, the recommendation for reactivity with canine samples is made based on 100% homology of the epitope of the antibody with the sequence for canine Vimentin.
