Vimentin Antibody (V9) is a mouse monoclonal IgG1 kappa light chain antibody raised against purified vimentin from porcine eye lens, a critical cytoskeletal protein primarily expressed in mesenchymal cells. Vimentin mouse monoclonal antibody (V9) recognizes an epitope within the vimentin immunogen sequence and demonstrates broad species cross-reactivity, including mouse, rat, and human samples, in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry on paraffin-embedded sections (IHCP), and flow cytometry (FCM) applications. Anti-vimentin monoclonal antibody (V9) is available in non-conjugated form and multiple conjugated forms including horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, agarose, and various Alexa Fluor® conjugates, offering versatility for diverse experimental needs. Vimentin belongs to the intermediate filament family and maintains cellular structure, integrity, and mechanical resilience. Vimentin interacts with numerous proteins including cytokeratins, actin, and microtubules to form an intricate network of cytoskeletal fibers. Vimentin′s association with motor proteins facilitates intracellular transport and organelle positioning. While predominantly cytoplasmic, vimentin localizes to the nucleus and cell membrane under specific conditions. Vimentin serves as a key marker for epithelial-to-mesenchymal transition (EMT), a process crucial in wound healing, development, and tumor metastasis. Vimentin′s expression pattern becomes particularly significant in cancer biology, as vimentin frequently appears upregulated in various neoplasms including melanoma, sarcomas, and breast carcinomas, where vimentin often shows co-expression with cytokeratins and other intermediate filaments. Vimentin (V9) antibody enables researchers to investigate cytoskeletal dynamics, cancer progression, and tissue-specific expression patterns through vimentin′s consistent cytoplasmic and membrane staining properties.

What is the KD (dissociation constant) of this antibody?

Thank you for your question. We do not measure or determine dissociation constant for our antibodies .We do know some are higher affinity than others ,but we so not measure or analyze this information.

I stained a cancer cell line with this mouse antibody and a rabbit one, but this didn't only work even at 1:50. Do you recommend a directly conjugated mouse monoclonal primary antibody, and why?

What is the epitope used for Vimentin V9 antibody? Is it the same as the old vimentin C20 or H84?

Thank you for your question. Vimentin Antibody (V9): sc-6260 has been raised against purified Vimentin from porcine eye lens, but its exact epitope region has not been determined, so its epitope is not known. We also offer Vimentin Antibody (E-5): sc-373717, which is specific amino acids 419-438 near the C-terminus of Vimentin of human origin.

Rated 5 out of 5 by from Great signal, highly specific and with fast timesWe tested the sample for the mouse monoclonal antibody for Vimentin from Santa Cruz Biotechnology. We are very pleased with the results, as the antibody performed exceptionally well in our experimental conditions. Nitrocellulose membrane with our total protein extracts transferred onto it was incubated for 1 hour with 5% milk solution in TBS-T, to block non-specific binding sites. Then, primary antibody sc-6260 was incubated for 2 hours at RT diluted 1:1000 in TBS-T with 0.02% Sodium Azide. Lastly, secondary anti-mouse antibody was incubated for 1 hour at RT diluted 1:2000 in 3% milk solution in TBS-T. HRP signal was detected with ECL incubation and quick film development. The signal appeared as a single specific band detected at around 57kDa, as indicated in the datasheet
