VCAM-1 Antibody (1G11) is a mouse monoclonal IgG1 kappa light chain antibody raised against the full-length human Vascular Cell Adhesion Molecule 1 (VCAM-1), also known as CD106. VCAM-1 is a transmembrane glycoprotein predominantly expressed on the surface of activated endothelial cells. Cellular localization on the membrane is crucial as VCAM-1 mediates leukocyte adhesion to the endothelium, facilitating transmigration during immune responses and inflammation. This cell-surface positioning allows VCAM-1 to interact directly with integrins like VLA-4 (α4β1) on leukocytes, playing a significant role in immune surveillance and inflammatory disease pathogenesis. Pro-inflammatory stimuli such as interleukin-1 (IL-1), tumor necrosis factor-alpha (TNF-α), and lipopolysaccharide (LPS) upregulate VCAM-1 expression, linking VCAM-1 to conditions like atherosclerosis, rheumatoid arthritis, and other vascular inflammatory disorders. Anti-VCAM-1 antibody (1G11) aids in studying endothelial activation, leukocyte adhesion, and mechanisms underlying vascular inflammation. VCAM-1 (1G11) antibody reacts with human VCAM-1 and is suitable for western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Understanding VCAM-1 cellular localization and function is essential for developing therapeutic strategies against diseases involving abnormal leukocyte trafficking and endothelial dysfunction.

How can VCAM-1 (1G11): sc-18854 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: Professor Griffin
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-18854 PE) or fluoroscein (sc-18854 FITC).
