    TTP Antibody (H-12) is a mouse monoclonal IgG2b antibody that detects tristetraprolin (TTP) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TTP is a crucial zinc-binding protein encoded by the immediate-early response gene Zfp-36, playing a significant role in the regulation of mRNA stability and inflammatory responses. TTP (H-12) antibody recognizes a protein essential for post-transcriptional regulation of pro-inflammatory cytokines, particularly tumor necrosis factor-alpha (TNF-α), which maintains immune homeostasis. TTP′s binding to AU-rich elements in the 3′ untranslated region of target mRNAs leads to their degradation, preventing excessive inflammation. Phosphorylation of TTP by p42 MAP kinase upon mitogen stimulation results in translocation from the nucleus to the cytoplasm, highlighting dynamic regulation in response to cellular signals. Studies using knockout mice deficient in TTP have revealed a propensity for developing autoimmune conditions, such as inflammatory arthritis and dermatitis, which can be mitigated by inhibiting TNF-α activity, underscoring TTP′s essential role in modulating inflammatory processes.     Hello, I recently used this Ab for WB with lung homogenates from experimental mice. Even though I saw a clear band, the size of the band was unusually high (above 100kDa). Could you give me an insight about why that could be the case?

    Thank you for your question. We do not have any reference or data supporting that high of a molecular weight for TTP. We would recommend that you include a positive control in your Western blot testing, such as 3T3-L1, or PC-12 whole cell lysates for comparison.     Has this antibody been shown to detect endogenous levels of TTP in mouse Raw 264.7 cells?

    Thank you for your question. We have not yet tested this antibody, TTP (H-12): sc-398904, in RAW 264.7 cells. However, we do have endogenous data in other mouse cell lines including RAW 309 Cr.1 and 3T3-L1 whole cell lysates.
