TSEN54 Antibody (A-9) is a mouse monoclonal IgG2b antibody that detects TSEN54 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TSEN54, also known as tRNA splicing endonuclease 54 homolog or SEN54, is a crucial 526 amino acid protein that plays a vital role in the tRNA splicing endonuclease complex, which is responsible for identifying and cleaving pre-tRNA at both 5′ and 3′ splice sites. This process is essential for the proper maturation of tRNA, as TSEN54 releases introns and generates free tRNA molecules with 2′,3′ cyclic phosphates and 5′-OH termini. Additionally, TSEN54 is involved in mRNA processing, linking pre-tRNA and pre-mRNA splicing events through association with pre-mRNA processing factors. Nuclear localization of TSEN54 allows for efficient processing of RNA molecules, which is critical for gene expression and cellular function. Defects in TSEN54 have been associated with pontocerebellar hypoplasia (PCH) types 4 and 2A, conditions characterized by structural abnormalities in the cerebellum, inferior olive, and ventral pons, highlighting TSEN54′s importance in neurodevelopment. TSEN54 exists as two alternatively spliced isoforms, further underscoring its complexity and functional diversity in cellular processes.
