Transferrin plays a crucial role in iron metabolism, serving as the primary iron transport protein in vertebrate serum. Transferrin is responsible for binding and transporting iron, which is essential for numerous biological processes, including oxygen transport, DNA synthesis, and cellular respiration. Transferrin consists of two homologous domains, each containing an iron-binding site, allowing effective iron sequestration and delivery to cells. While the liver is the main site of transferrin synthesis, transferrin is also produced in smaller quantities in the testis and brain, as well as in oligodendrocytes, where transferrin serves as an early marker of oligodendrocyte differentiation. Transferrin binds to the transferrin receptor (TfR), also known as CD71, on the surface of erythroblasts and reticulocytes, facilitating iron uptake through a process involving coated pits and vesicles. Following iron release, transferrin is recycled back into the extracellular space for reuse. Deficiencies or mutations in the transferrin gene can lead to atransferrinemia, a rare autosomal recessive disorder characterized by microcytic anemia and iron overload, highlighting transferrin′s importance in maintaining iron homeostasis in the body.
