TPN Antibody (E-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects TPN protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TPN monoclonal antibody (E-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TPN protein, also known as tapasin, TPSN, TAPBP, or transporter associated with antigen processing-A, is a type I membrane glycoprotein encoded by a gene located on chromosome 6p21, producing a 488-residue protein. One of the critical aspects of TPN′s function is its role in the assembly and stabilization of major histocompatibility complex (MHC) class I molecules. TPN facilitates the loading of peptide antigens onto MHC class I molecules within the endoplasmic reticulum, a process essential for effective antigen presentation to CD8+ T cells. The cytoplasmic domain of TPN contains a double-lysine motif that is crucial for retention in the endoplasmic reticulum, ensuring that MHC class I molecules are properly loaded with peptides before being transported to the cell surface. Deficiencies in TPN lead to impaired MHC class I expression and reduced immune responses, highlighting TPN′s importance in the immune system′s ability to recognize and respond to intracellular pathogens.

Can TPN (E-11): sc-393552 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, TPN (E-11): sc-393552 has been validated for use in IHC with paraffin-embedded sections.
