TPN Antibody (TO-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects TPN in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). TPN Antibody (TO-3) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TPN, also known as tapasin, TPSN, TAPBP, or transporter associated with antigen processing-A, is a type I membrane glycoprotein encoded by a gene located on chromosome 6p21, producing a 488-residue protein. TPN plays a crucial role in the immune response by facilitating the transfer of peptide antigens onto major histocompatibility complex (MHC) class I molecules, which is essential for the presentation of antigens to CD8+ T cells. The cytoplasmic domain of TPN contains a double-lysine motif that is critical for retention in the endoplasmic reticulum, ensuring newly assembled MHC class I molecules are properly loaded with antigens before transport to the cell surface. TPN knockout mice exhibit significant defects in MHC class I expression and antigen presentation, underscoring TPN′s importance in maintaining effective immune responses.

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-80647 PE) or fluoroscein (sc-80647 FITC).
