TLE4 Antibody (E-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects TLE4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TLE4 (E-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TLE4, also known as Groucho-related protein 4, plays a crucial role in the Notch signaling pathway, which regulates cellular interactions that determine cell fate during development in both invertebrates and vertebrates. TLE4 functions as a transcriptional repressor, interacting with chromatin and specifically with Histone H3, thereby influencing gene expression patterns critical for processes such as neurogenesis and epithelial differentiation. TLE4 is predominantly expressed in the brain, particularly in the caudate nucleus, and expression levels correlate with the differentiation status of epithelial cells. The dynamic regulation of TLE4, along with family members TLE1, TLE2, and TLE3, highlights importance in cellular differentiation, as TLE1 is upregulated in early differentiation stages while TLE2 and TLE4 are upregulated later, indicating roles in the transition of cells toward a terminally differentiated state. This intricate regulation underscores TLE4′s significance in developmental biology and potential implications in understanding various pathologies, including cancers where TLE family members exhibit altered expression patterns.

Rated 1 out of 5 by from Not working on human post-mortex prefrontal cortexI bought this antibody a few weeks ago, I tested it on human post-mortem FFPE prefrontal cortex tissue, with very scarse results. The sections were dewaxed with 2x5’ xylene, rehydrated with 2x5’ 100%, 5’ 90% and 5’ 70% EtOH and washed with dH2O 2x10’. Antigen retrieval was performed with EDTA pH9 20’ at 95C. Sections were then let cool down 10’ in ice, then washed 10’ in dH2O and 10’ in PBS. Blocking was performed with blocking buffer (10% donkey serum in PBS with 0.3%Triton and 0.003% Sodium Azide) for 1hr at RT. Anti-TLE4 antibody was diluted 1:200 in blocking buffer and incubated at 4C for 1 night. Anti-mouse Alexa 647 secondary antibody was then applied after 3x 5’ washed in PBS, diluted 1:500 in blocking buffer with DAPI, for 1 hr at RT. Sections were then washed 3x 5’ in PBS, stained with Sudan black for 10’, washed again in dH2O, air dried and mounted. Confocal acquistion of the staining is attached: TLE4 seems to stain most of nuclei, with no layer specificity. I can't use this antibody for my studies and I would like to get a refund.
