TIGAR Antibody (E-2) is a mouse monoclonal IgG2b kappa light chain antibody that detects TIGAR protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). TIGAR (E-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TIGAR protein, also known as TP53 (tumor protein 53)-induced glycolysis and apoptosis regulator or C12orf5, is a 270 amino acid protein that plays a crucial role in cellular metabolism and stress response. Induced by the p53 tumor suppressor pathway, TIGAR functions to mitigate oxidative stress by modulating glycolytic flux. Specifically, TIGAR shares sequence similarity with the bisphosphate domain of fructose-2,6-bisphosphate degrading enzyme, allowing TIGAR to lower intracellular levels of fructose-2,6-bisphosphate. By inhibiting glycolysis, TIGAR redirects metabolic pathways toward the pentose phosphate shunt, which is vital for generating NADPH and ribose-5-phosphate, essential for nucleotide synthesis and cellular antioxidant defense. This mechanism underscores TIGAR′s importance in protecting cells from reactive oxygen species, which can cause DNA damage and trigger apoptosis, thereby highlighting TIGAR′s potential role in cancer biology and therapeutic strategies.

I am using TIGAR (E-2): sc-166290 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
