TFR2 Antibody (9F8 1C11) is a mouse monoclonal IgG1 kappa light chain antibody that detects TFR2 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-TFR2 antibody (9F8 1C11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The transferrin receptor 2 (TFR2) plays a crucial role in iron metabolism by mediating the uptake of transferrin-bound iron, which is essential for numerous biological processes, including oxygen transport and DNA synthesis. TFR2 is predominantly expressed in the liver, particularly in hepatocytes and erythroid precursors, highlighting its importance in maintaining iron homeostasis and supporting erythrocyte differentiation. The gene encoding human TFR2 is located on chromosome 7q22 and produces two isoforms: an α isoform that encodes a transmembrane protein and a β isoform that encodes a shorter, intracellular protein. Dysregulation or mutations in TFR2 can lead to hereditary hemochromatosis type III (HFE3), a serious iron overload disorder associated with significant clinical complications such as cirrhosis, cardiomyopathy, diabetes, endocrine dysfunctions, arthropathy, and an increased risk of liver cancer. Using TFR2 monoclonal antibody (9F8 1C11) in research can provide valuable insights into iron metabolism mechanisms and the pathophysiology of related disorders.

Thank you for your question. The immunogen used for this TFR2 antibody, does not show significant homology with the rat or mouse sequence for TFR2. At this time our TFR2 antibodies are not recommend for detection of TFR2 in rats or mouse.

How much protein do you recommend loading on the gel for Western blot detection using TFR2 (9F8 1C11): sc-32271 monoclonal antibody?

Asked by: jenniferc
We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
