  • To inhibit serine/threonine phosphatase activity, add 50 mM Sodium Flouride (NaF) (sc-24988) to Blotto. Alternatively, add Phosphatase Inhibitor Cocktail A (sc-45044) and Phosphatase Inhibitor Cocktail B (sc-45045)
  • Recommended to reduce overall background, and minimize non-specific signals commonly associated with enhanced chemiluminescent (ECL) protein detection methods using HRP conjugated primary and secondary antibodies, or mouse IgG binding proteins
  • If using direct conjugated primary antibodies or for near-infrared (NIR) detection methods, we strongly recommend our alternative product, UltraCruz® Blocking Reagent (sc-516214)
  • If not blotting with phospho-specific antibodies, we recommend our alternative product, TBS Blotto A (sc-2333) which contains a unique formulation of 5% milk, 0.05% Tween-20

TBS Blotto B is a blocking solution commonly used in Western blotting and other immunoassay techniques, similar to TBS Blotto A. It typically contains Tris-buffered saline (TBS), which consists of Tris-HCl and NaCl, along with a blocking agent such as non-fat dry milk or bovine serum albumin (BSA). In Western blotting, TBS Blotto B serves the same purpose as TBS Blotto A: to block non-specific binding sites on the membrane, thereby reducing background noise and increasing the specificity and sensitivity of antibody detection. TBS Blotto B is added to the membrane after the primary antibody incubation step and before the secondary antibody incubation step in Western blotting protocols. It is incubated with the membrane for a period of time to ensure complete blocking of non-specific binding sites. Similarly to TBS Blotto A, TBS Blotto B can also be used as a diluent for primary and secondary antibodies, as well as for washing steps in immunoassay procedures. Its composition provides a suitable environment for antibody-antigen interactions and helps maintain the stability of proteins during the assay.
