Tau Antibody (D-8) is a mouse monoclonal IgG2b kappa light chain antibody designed to recognize Tau protein, also known as MAPT (microtubule-associated protein tau), or TAU. Tau monoclonal antibody (D-8) targets the amino acids 1-150 region of the human Tau protein and is effective in detecting Tau from mouse, rat, and human sources using techniques such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Tau is a 758 amino acid protein predominantly expressed in neuronal tissues, where Tau plays a crucial role in promoting microtubule assembly and stability, maintaining neuronal polarity, and facilitating cytoskeletal plasticity by linking microtubules to plasma membrane components. Tau protein exists in multiple alternatively spliced isoforms and is localized to the cytoplasm, cytoskeleton, and cell membrane. Tau undergoes several post-translational modifications, such as phosphorylation on serine and threonine residues, polyubiquitination leading to proteasomal degradation, and glycation of specific isoforms. These modifications are essential for regulating Tau′s binding to microtubules, degradation, and ability to form aggregates, which are critical factors in normal function and involvement in disease processes. Dysregulation of these post-translational modifications is associated with neurodegenerative disorders, including Alzheimer′s disease, pallido-ponto-nigral degeneration (PPND), corticobasal degeneration (CBD), and progressive supranuclear palsy (PSP).
